Meridian’s Air-Dryable ™ DNA LAMP and RNA/DNA LAMP Mixes are ready-to-use, 4x concentrated, fully optimized mixes for isothermal amplification methods such as LAMP, designed to dramatically speed up assay development time and reduce TTR by 50%. Isothermal amplification is a highly sensitive nucleic acid amplification method that has gained popularity in recent years due to its advantages over qPCR, such as quicker time to results, simple assay workflow, and point-of-care usability. However, the initial assay design and development can be more complex with 4 to 6 primers which are used in combination to generate a robust, fast and sensitive result. Through extensive R&D research, Meridian has discovered a blend of enzymes, dNTPs, Mg 2+ , excipients and stabilizers that are used with customer-selected primers and probes for creating any custom DNA or RNA LAMP assay. Experimental data on both RNA and DNA templates demonstrate that the mixes are up to 2x faster than other suppliers, generating results in half the time. In addition, the mixes incorporate Meridian’s new Air-Dryable ™ technology and can be used in a liquid format or oven-dried to create an ambient-temperature stable assay (please see the product guide and FAQs for recommendations on oven drying parameters).

Air-Dryable ™ RNA/DNA LAMP mix was air-dried with Dengue type 2 primers at 50°C for 80 mins. The sensitivity of air-dried Air-Dryable ™ RNA/DNA LAMP mix ( red ) was compared to its liquid ( blue ) format and NEB WarmStart ® ( orange ) in a serial dilution of the target RNA (Dengue type 2 crude lysate). Reactions were incubated at 65°C for 60 min and TTR were measured at 1:10 of end fluorescence. Error bars represent standard deviation over 4 technical replicates. The results illustrate that the speed of Air-Dryable ™ RNA/DNA LAMP mix, is twice as fast as NEB WarmStart ® in either the dry or liquid format.

The average time to results (TTR) of Air-Dryable ™ LAMP Mixes in both air-dried (red) and liquid (blue) formats vs. NEB WarmStart ® (orange). Results demonstrate greater sensitivity of Air-Dryable ™ LAMP Mixes with shorter TTR across all template concentrations. (A) Conserved regions from the S and P genes of Hepatitis B Virus (1000, 100 and 20 copies respectively) were amplified using Air-Dryable ™ DNA LAMP (MDX119) in an air-dried ( red ) or liquid ( blue ) format or using NEB WarmStart ® ( orange ). Reactions were incubated at 65°C for 60 min and TTR were measured at 1:10 of end fluorescence. At lower copy numbers, NEB WarmStart ® ( orange ) exhibited significantly lower reaction reproducibility compared to Air-Dryable ™ DNA LAMP Mix, and a lower TTR was observed for reactions using Air-Dryable ™ DNA LAMP Mix across all dilutions of the Hepatitis B gene. (B) Emesvirus (MS2) was amplified using Air-Dryable ™ RNA/DNA LAMP (MDX118) in an air-dried ( red ) or liquid ( blue ) format or using NEB WarmStart ® ( orange ). Reactions were incubated at 65°C for 60 min and TTR were measured at 1:10 of end fluorescence. Error bars represent standard deviation over 4 technical replicates. A significantly lower TTR was observed for reactions using Air-Dryable ™ DNA/RNA LAMP Mix across all dilutions of MS2-RNA.

High reaction specificity with little-to-no non-specific amplification

Air-Dryable ™ RNA/DNA LAMP (MDX118) ( blue ) was compared to NEB WarmStart ® ( orange ) for detection of EBV DNA (1000 ipc) and MS2 RNA (1000 ipc). Reactions were incubated at 65°C for 60 min and TTR were measured at 1:10 of end fluorescence. Amplification plots of 3 technical replicates show Air-Dryable ™ RNA/DNA LAMP mix has earlier TTR and no non-specific amplification compared to NEB WarmStart ® which has detectable amplification for non-template controls (NTC).

Air-Dryable ™ DNA LAMP and RNA/DNA LAMP mixes were air-dried and their stability was tested in an accelerated stability study. A) 9TV10 from Trichomonas vaginalis was amplified with Air-Dryable ™ DNA LAMP and B) Dengue Type 2 was amplified with Air-Dryable ™ RNA/DNA LAMP that was air-dried ( red ) and incubated a 37°C for over 1 month and tested against the fresh liquid mix ( blue ). Results suggest that the air-dried mixes retain full activity following accelerated stability tests with a projected stability of greater than 12 months at ambient temperature.
