Anticoagulants and high concentrations of whole blood are known to inhibit efficiency of qPCR reactions. Whole human blood at concentrations of 2%, 5%, and 10% were tested in the presence of K2-EDTA ( orange ), sodium heparin ( green ), and sodium citrate ( grey ). These tests were conducted using A) Lyo-Ready AOF dUTP DNA qPCR Blood (MDX322) and compared with Roche KAPA Probe Force qPCR Master Mix and ThermoFisher TaqPath ™ ProAmp ™ Multiplex Master Mix and B) Lyo-Ready AOF dUTP RNA/DNA qPCR Blood (MDX323) and compared with Quantabio UltraPlex ™ 1-Step ToughMix ® , and ThermoFisher TaqPath ™ 1-Step Multiplex Master Mix. The results demonstrate that Lyo-Ready AOF dUTP Blood mixes maintain higher performance and consistently earlier Ct values in the presence of inhibitors in high concentrations of blood containing common anticoagulants, ensuring reliable amplification even under challenging conditions.
