DNA polymerase activity of lyophilized Glycerol-Free Phi 29 DNA Polymerase (HC) ( red ) was measured in a rolling circle amplification (RCA) assay, to amplify single stranded circular microphage DNA, and compared against Φ 29 DNA Polymerase (high concentration) from Qiagen ( black ). The results demonstrate that Glycerol-Free Phi 29 DNA Polymerase (HC) has greater DNA polymerization activity than the product from other suppliers.

Lyophilized Glycerol-Free Phi29 (HC) Maintains Full Activity
Glycerol-Free Phi29 DNA Polymerase (HC) (MDX240) was lyophilized in its reaction buffer, and its stability was tested in an accelerated study. The lyophilized enzyme ( red ) was incubated at 37°C for one month, its DNA polymerization activity was tested in RCA assay against a liquid aliquot stored at -80°C ( blue ). The results show that lyophilized MDX240 remains active, with projected stability of up to 12 months at ambient temperature.

Glycerol-Free Phi29 (HC) Remains Stable After Freeze-Thaw Cycles
Glycerol is a cryoprotectant and is normally part of the storage buffer, where it serves to protect Phi 29 DNA Polymerase during freezing conditions. To test the resistance of Glycerol-Free Phi 29 DNA Polymerase (HC) to freezing and thawing, a freeze‑thaw test was performed ( 5 cycles and 10 cycles of freeze/thawing) and compared to fresh product ( 0 cycles of freeze/thawing). The enzyme was tested using RCA to amplify single stranded circular microphage DNA. The results illustrate that Glycerol-Free Phi 29 DNA Polymerase (HC) is stable, showing the same performance before and after multiple rounds of freeze/thawing.
