More confidence when low-copy targets matter. Residual E. coli DNA associated with recombinant enzymes produced in bacterial systems can create background signal and increase false-positive risk. Meridian non-bacterial (Non-Bac) qPCR solutions use a eukaryotic expression system to support sensitive detection of low-copy bacterial targets.

MDX362 Performance Data

Designed for Sensitive Bacterial DNA Detection The completed 4x hot-start mix containing optimized buffer chemistry, PCR enhancers, dNTPs (including dUTP), MgCl 2 and Glycerol-Free Non-Bac HS Taq polymerase, supports low-copy and multiplex qPCR testing. It can be used as a liquid mix or lyophilized for ambient-stable assay formats.

Lyo-Ready Non-Bac DNA qPCR Mix in liquid (blue) and lyophilized (red) format were compared for their ability to amplify a 10-fold serial dilution of bacterial DNA target (10,000, 1,000, 100, 10 and 1 copies respectively). The results illustrate that the lyophilized mix maintains the same high application efficiency as the liquid mix, with high sensitivity and reproducibility. NTCs are shown in (black) .

No Detectable Bacterial DNA Contamination

Using a pan-bacterial 16S assay, MDX362 showed high sensitivity and reproducibility while the no-template control remained without detectable amplification. Lyo-Ready Non-Bac DNA qPCR Mix was used to amplify a 10-fold serial dilution of bacterial DNA target (10,000, 1,000, 100, 10, and 1 copies, respectively) in liquid (blue) format using a pan-bacterial 16S assay. No detectable amplification was observed in the no-template control (black) . The Lyo-Ready Non-Bac DNA qPCR Mix demonstrates high

sensitivity and reproducibility, supporting reliable detection of low-copy bacterial DNA targets with no detectable bacterial background amplification under the conditions tested.

Three bacterial DNA targets were amplified using Lyo-Ready Non-Bac DNA qPCR Mix in liquid (blue) format, under standard qPCR cycling conditions. The results demonstrate the high sensitivity of Lyo-Ready Non-Bac DNA qPCR Mix, enabling reliable amplification of bacterial targets down to single-copy levels in multiplex reactions. NTCs are shown in (black) .

Lyophilized Assay Stability Lyo-Ready Non-Bac DNA qPCR Mix was lyophilized, and the stability of the dried assays (red) was tested after accelerated stability against fresh liquid mix (blue) in a triplex qPCR assay. The lyophilized mix retained performance comparable to the liquid mix, supporting projected stability of more than 12 months at ambient temperature.

The activity of the Glycerol-Free Non-Bac HS Taq Polymerase (HC) was tested in a triplex qPCR assay targeting marker genes from three bacteria (16S rRNA gene from Enterococcus , Mip gene from Legionella pneumophila and ToxA gene from Pseudomonas aeruginosa ), using a 10-fold serial dilution of the bacterial genomic DNA. The result shows that Glycerol-Free Non-Bac HS Taq Polymerase (HC) can successfully detect multiple targets down to 1 copy of DNA in multiplex assays.

Lyophilized Assay Stability Glycerol-Free Non-Bac HS Taq Polymerase (HC) was lyophilized in a lyo-ready DNA qPCR mix and the stability of the dried assay (red) was tested after accelerated stability against fresh liquid mix (blue) in a triplex qPCR assay. The lyophilized mixes retained the same performance as the liquid. The lyophilized mix retained performance comparable to the liquid mix, supporting projected stability of more than 12 months at ambient temperature.
