Glycerol-Free Taq HS is an antibody hot-start Taq DNA polymerase designed for developing lyophilized assays. The highly concentrated (50 U/µL) glycerol-free solution, is ideal for the preparation of dried-down amplification mixtures intended for high-throughput assay platforms. It is supplied as a separate taq, antibody and enzyme dilution buffer (all glycerol-free) to provide greater flexibility in assay development.

Glycerol is a cryoprotectant and is normally part of the storage buffer, where it serves to protect Taq polymerase during freezing conditions. To test the resistance of Glycerol‑Free Taq Hot‑Start to freezing and thawing, a freeze‑thaw test was performed (10 and 15 cycles of freeze/thawing) and compared to fresh product (0 cycles of freeze/thawing). The results illustrate that Glycerol‑Free Taq Hot‑Start offers the same protection to freeze/thawing as a polymerase that contains glycerol.

Comparison of freshly prepared and lyophilized glycerol-free Taq HS master mixes
A 10-fold serial dilution of template DNA was used to set up two sets of qPCR assays. One set was lyophilized (blue) and the other left as a wet mix (black). Results illustrate that lyophilization of Glycerol-Free Taq HS does not have an effect on the quality of the qPCR, as both the assays demonstrate identical efficiency and sensitivity.
